A greater understanding of factors influencing fertility is essential to improve pregnancy rates and reduce the occurrence of embryonic mortality in beef herds. The objective of the current study was to evaluate retrospective data of pregnancy per artificial insemination (P/AI) and pregnancy loss in Nelore females subjected to timed-AI (TAI) in Brazil. Data from 40,104 TAI collected from six breeding seasons (2016-2022) were analyzed, and the effects of animal category (e.g., classification based on age and parity), farm, month of parturition, sire, sire breed (Nelore vs Angus), estrus expression at TAI, animal temperament, and body condition scores (BCS) were evaluated. P/AI and pregnancy loss were affected (P < 0.001) by animal category. There was also an effect of farm (P = 0.0013) on P/AI and pregnancy loss (P = 0.001), as P/AI ranged from 49.28% and 55.58% and pregnancy loss from 3.37% to 6.89% across the herds evaluated. Month of parturition also affected (P < 0.001) P/AI and was higher for cows that became pregnant at the beginning of the previous breeding season. Calmer animals, presenting lower velocity scores while exiting the chute following TAI, achieved higher P/AI (P < 0.001). Lower BCS at TAI was associated (P < 0.001) with increased pregnancy loss, and BCS gain following AI was associated (P < 0.001) with reduced rates of embryonic mortality. There was a major effect (P < 0.001) of sire on P/AI and pregnancy loss, as P/AI ranged from 11% to 79%, and embryonic mortality from 0% to 40% for the bulls used in the study, highlighting the importance of the sire fertility on overall pregnancy success. Results from the current study reinforce the idea that animal age and parity at the beginning of the breeding season, BCS at the onset of estrous synchronization, BCS gain following AI, estrus expression at TAI, sire, and month of parturition are important factors influencing P/AI and rates of embryonic mortality in beef herds.

Despite being the most important form of biotechnology in animal reproduction, artificial insemination was used in about 23% of Brazilian bovine herds in 2021. This is due to the variability of results caused by varying bull fertility and body condition of the cows. This study aimed to correlate the fertility indices of bulls with qualitative attributes of the semen. Semen samples from 28 bulls (Nellore and Angus) were used to evaluate postthaw sperm morphology and kinetics using conventional analysis, image-based flow cytometry (IBFC) and computer-assisted semen analysis (CASA). The fertility index was effective in separating bulls into 4 different fertility classes (P < 0.001), and fertility rates in timed artificial insemination (TAI) remained constant between the cows' fertility categories (P < 0.001) and in the different grades of female body condition (P < 0.005). After partial least squares regression (PLS) analysis, four models were proposed with different variables. The coefficients of determination for the conventional analysis, CASA, and IBCFC models were 0.154, 0.380, and 0.259, respectively. The composite model, including select IBFC and CASA parameters, showed a greater R2 (0.481) with progressive motility, average speed (VAP, µm/s), membrane integrity, and mitochondrial potential, showing a positive effect. Linear speed (VSL, µm/s) and acrosomal integrity had a negative effect on bull fertility indices. Bulls classified by the fertility index attained dispersed pregnancy rates in different cow body condition score (BCS) classes, and the sperm quality pattern was consistent with this classification. In conclusion, this novel composite model including CASA and IBFC parameters improves the prediction of bull fertility used in TAI.

This study is part of a concerted effort to identify and phenotype rare, deleterious mutations that adversely affect sperm quality, or convey high developmental and fertility potential to embryos and ensuing progeny. A rare, homozygous mutation in EML5 (EML5 R1654W ), which encodes a microtubule-associated protein with high expression in testis and brain was identified in an Angus bull used extensively in artificial insemination (AI) for its outstanding progeny production traits. The bull's fertility was low in cross-breeding timed AI (TAI) (Pregnancy/TAI = 25.2%; n = 222) and, in general, AI breeding to Nellore cows (41%; n = 822). A search of the 1,000 Bull Genomes Run9 database revealed an additional 74 heterozygous animals and 8 homozygous animals harboring this exact mutation across several different breeds (0.7% frequency within the 6,191 sequenced animals). Phenotypically, spermatozoa from the homozygous Angus bull displayed prominent piriform and tapered heads, and outwardly protruding knobbed acrosomes. Additionally, an increased retention of EML5 was also observed in the sperm head of both homozygous and heterozygous Angus bulls compared to wild-type animals. This non-synonymous point mutation is located within a WD40 signaling domain repeat of EML5 and is predicted to be detrimental to overall protein function by genomic single nucleotide polymorphism (SNP) analysis and protein modeling. Future work will examine how this rare mutation affects field AI fertility and will characterize the role of EML5 in spermatogenesis.

The finishing stages for uncastrated Nellore beef cattle on tropical pasture often produce high yielding carcasses with minimal fat cover. This study aimed to evaluate the performance and carcass characteristics of uncastrated Nellore beef bulls fed a diet containing rumen-protected fat during the rainy period for 210 days. Eighty Nellore beef bulls with 28 months of age, and an initial live weight of 416 kg ± 16 kg, were allocated into eight paddocks (7.0 ha), totaling 56 ha, at 10 animals/paddock, in a continuous-grazing system. The pasture characteristics, performance production, loin-eye area (cm2), subcutaneous fat thickness (mm), and rump fat thickness were evaluated. There was no effect (P > 0.05) on the average total weight gain (203.00 kg) and average daily gain (0.98 kg/day). On the other hand, the subcutaneous fat thickness and rump fat thickness were 42.8% and 49.5% greater, respectively, when the animals were fed a diet containing rumen-protected fat inclusion (P < 0.05). Supplementation with rumen-protected fat provided a thicker fat layer in the carcass of Nellore beef bulls grazing on tropical pasture during the rainy period.

We hypothesized that by-products from biodiesel production like crambe meal (Crambe abyssinica) totally replace soybean meal (Glycine max) and crude glycerin could partially replace corn (Zea mays). Our aim was to evaluate different supplements with combinations of crambe meal and crude glycerin replacing soybean meal or corn on performance and carcass traits of finishing heifers. Sixty Nellore heifers 18 months old were used, and 298.96 ± 21.99 kg of body weight allotted in Brachiaria brizantha pastures. All heifers were distributed in a randomized complete block design, with four treatments being the following supplements: corn + soybean meal (CS), corn + crambe meal (CCr), corn + soybean meal + crude glycerin (CSG), corn + crambe meal + crude glycerin + (CCrG). The animals were distributed in four paddocks of 13 hectares each, in a continuous grazing system. We evaluated pasture characteristics, intake, performance, carcass traits, characteristics, and centesimal composition of Longissimus thoracis muscle. There was an effect (P < 0.05) of inclusion of crambe meal on the performance of the animals for slaughter weight and daily gain. The supplement containing corn + soybean meal (CS) presented better conversion rates and food efficiency. There was an effect of inclusion of crambe meal and crude glycerin on the centesimal composition of Longissimus thoracis muscle. The use of crambe meal and crude glycerin as alternative sources from the agro-industry of biodiesel in concentrate supplement for finishing heifers in pastures can promote positive effects related to productive performance and carcass characteristics.

The aim of this study was to evaluate the impact of increased shadow supply in integrated crop-livestock-forest systems on in vitro embryonic development and physiological parameters related to stress response in Nellore heifers (Bos indicus). For the study, animals (n = 16) were randomly divided into two groups and kept in areas with different afforestation systems, the integrated crop-livestock-forest (ICLF) and the integrated crop-livestock (ICL) system. The microclimate of the ICLF system provided better comfort conditions than ICL. No differences of respiratory rate, rectal temperature, cortisol, T3, T4, oocyte quality, and cleavage rate between the systems were verified. A higher blastocyst rate was observed in the ICLF (p < 0.05). The results demonstrate that Nellore heifers managed in ICLF during summer in Midwest of Brazil showed higher production of in vitro embryos, without typical changes in its physiological parameters. The results observed in the present study indicate that zebu females are able to respond satisfactorily to the intense heat conditions; however, we believe that the long period to which these animals are exposed to these conditions interferes in the oocyte competence and embryo development.

Due to the increasing use of in vitro embryo production (IVEP) and the importance of crossbreeding for beef production, pregnancy rates of Nelore recipients were evaluated following Fixed Time Embryo Transfer with fresh or vitrified IVEP embryos produced with Y-sorted sperm of Angus bulls (B. taurus) or Fixed Time Artificial Insemination using non-sorted sperm. For IVEP in Experiment 1, oocytes were obtained using Ovum Pick Up (OPU) (n = 84 embryos) or from ovaries from a slaughterhouse (SLAUGHTER, n = 66 embryos). In Experiment 2, with oocytes obtained by OPU, IVEP embryos were fresh (FRESH, n = 271) or after vitrification/warming (VITRIFIED, n = 79) and PR was compared with FTAI (n = 239). In Experiment 1, cleavage rates were 63.8% and 39.1% for OPU and SLAUGHTER groups, respectively (P = 0.02), and blastocyst rates were 30.5% and 14.7%, respectively (P = 0.09). The PR was similar when considering the source of oocytes (OPU = 35.7%; SLAUGHTER = 25.8%; P = 0.17). In Experiment 2, there was no difference in PR for FRESH or VITRIFIED embryos (34.3% and 30.4%, respectively, P = 0.72), but lesser than FTAI (47.7, P = 0.002). It is concluded that the IVEP with Y-sorted sperm associated with vitrification or embryos produced with oocytes from different sources did not affect PR when there was transfer of crossbred embryos into recipients, and can optimize large-scale application of IVEP technology; however, FTAI pregnancy rates with non-sex sorted sperm were greater.

Bovine livestock is a major anthropogenic greenhouse gas source via enteric methane. Brazilian bovine livestock is also responsible for emissions from land-use changes. In contrast, enteric emissions from extensive cattle systems in wetlands might have been overestimated. We provide scientific evidences that the human footprint of bovine products delivered by the Pantanal can be much lower. To assess this, a historical cloud-free imagery of the Landsat-5, spanning 26 years, were processed for mapping spatiotemporal landscapes in a Pantanal farm under cattle intensification studies. Eight landscape categories were identified according to spatiotemporal dynamics of interannual floods. The spatiotemporal map allowed in the field the adoption of stratified random samplings of chamber gas fluxes. The combination of stratified sampled landscapes with Monte Carlo simulations of measured methane emissions in wet and dry soils permitted to integrate landscapes emissions at annual basis with biased uncertainties. Assuming enteric emissions obtained for the Pantanal region, our results suggest that the landscapes methane emissions are 10- to 23-fold superior than the enteric emissions of traditional bovine systems. While enteric emissions seem negligible with respect to net farmland emissions, cattle livestock provide important environmental services like carbon recycling through non-competing herbivory. Moreover, cattle might be making use of a biomass that would undergo decomposition during the flooding phase. Our analysis thus indicate that enteric emissions from traditional bovine systems in flooding farmlands could be considered neutral. By contrast, intensification to improve the stocking rate should be accounted as net anthropogenic emissions. A case study of intensification allowed an increase of 48% in the stocking rate, which is associated with net anthropogenic emissions from 534 bovine animals or about 27 to 63 Mg of enteric CH4 per year. In short, the competition between traditional and distinct levels of cattle intensification will result from a trade-off between public policies and strategic market niches (organic, sustainable) for the optimal landscape management of the Pantanal.

ABSTRACT: This study evaluated the reproductive performance of Nellore heifers without previous vaccination undergoing fixed-time artificial insemination (FTAI) and subsequent natural breeding under different vaccination protocols against infectious bovine herpesvirus 1 (BHV-1), bovine viral diarrhea virus (BVDV), and bovine leptospirosis (BL). A total of 763 animals were included; the group assignment was as follows: T1, control without vaccination; T2, vaccination against BL; T3, vaccination against BHV-1and BVDV; T4,vaccination against combination of BL + BHV-1and BVDV. Vaccination was started 30 days before FTAI (D-40), and a booster dose was administered at D-10. Serological tests were used to verify the occurrence rate of natural infection by BL, BHV-1, and BVDV. Initial pregnancy was diagnosed at D30, D60, and D120 after FTAI. Final pregnancy rates were evaluated by group, weight, body-condition score, occurrence of estrus, inseminator, bull, and occurrence of abortions or embryonic death. In all groups, results indicated seropositivity of 100% for leptospirosis, 77.0-94.0% for BHV-1, and 64.0-87.0% for BVDV. The vaccination protocol was not a significant influencing factor of pregnancy rate and embryonic or fetal mortality; whereas, animal weight at the beginning of the breeding season and presence of estrus at FTAI were significant influencing factors of pregnancy rate. The vaccination protocol had no interference with the reproductive performance of the experimental groups.


RESUMO: O estudo foi conduzido para avaliar o desempenho reprodutivo de novilhas Nelore, não imunizadas para herpesvirus bovino tipo 1 (BHV-1), vírus da diarreia viral bovina (BVDV) e leptospirose bovina (LB), submetidas a diferentes protocolos vacinais em manejo de inseminação artificial em tempo fixo (IATF) e repasse com touro em monta natural. Um total de 763 animais foram distribuídos em quatro grupos: T1 - controle sem vacinação; T2 - vacinação contra LB; T3 - vacinação contra BHV-1 e BVDV T4 - vacinação com LB+ BHV-1 e BVDV. A vacinação foi iniciada 30 dias antes da IATF (D-40) com a aplicação da segunda dose em D-10. Verificou-se a taxa de ocorrência de infecção natural por LB, BHV-1 e BVDV realizando-se exames sorológicos. O diagnóstico de gestação inicial foi feito aos 30, 60 e 120 dias. Avaliou-se as taxas de prenhez em função dos grupos, do peso, escore de condição corporal, presença ou ausência de cio, inseminador, touro, ocorrência de abortos ou morte embrionária. Os resultados obtidos revelaram que soropositividade para leptospirose foi de 100% para todos os grupos, variando de 77,0% a 94,0% para BHV-1 e 64,0% a 87,0% para BVDV. O protocolo vacinal não interferiu na taxa de prenhez, de mortalidade embrionária ou fetal. Houve diferença na taxa de prenhez em relação ao peso dos animais no início da estação reprodutiva e a presença de cio no momento da IATF. Concluiu-se que o protocolo vacinal não interferiu no desempenho reprodutivo dos grupos experimentais.

The objective of this study was to evaluate the effect of a PGF2α-analogue (PGF) on ovulation and pregnancy rates after timed artificial insemination (TAI) in cattle. In Experiment 1 cows received an intravaginal progesterone-releasing device (CIDR) plus 2 mg im of estradiol benzoate (EB) on Day 0. The CIDR devices were removed on Day 8, and all cows received 150 µg im of d-cloprostenol (PGF2α-analogue), 300 IU of eCG and 1 mg of estradiol cypionate (ECP) im. On Day 9, cows were randomly assigned into two groups: 1) ECP Group (n = 17), that did not receive any further treatment; and 2) ECP-PG Group (n = 14) that were given 150 µg of d-cloprostenol (PGF) as adjuvant stimulus for ovulation. No difference between groups was detected in interval for ovulation (P = 0.5), and in the proportion of cows ovulating (P = 0.09). In Experiment 2, multiparous suckling crossbred Aberdeen Angus cows (n = 260), were treated into two groups, similarly as Experiment 1; ECP group (n = 122), and ECP-PG group (n = 138). All females were TAI on Day 10. The proportion of cows treated with ECP that became pregnant was 54.9% (67/122), and cows treated with ECP plus PGF was 55.1% (76/138; P = 0.9). In Experiment 3, 686 Nelore cows, 40 to 50 days postpartum, were treated as Experiment 1 (ECP group), however, on Day 8 cows were divided into 3 groups: ECP Group (n = 216); PGF-SC Group (n = 228), in which cows did not receive ECP and were given an additional subcutaneous injection of PGF on Day 8; and PGF-IM Group (n = 242), in which cows also did not receive ECP on Day 8 and were given an additional injection of PGF im on Day 9. On Day 10, estrus was evaluated at timed AI (TAI). There was no difference in the diameter of the dominant follicle at CIDR removal and at TAI, and pregnancy per AI among groups (P > 0.05). However, the proportion of cows that displayed estrus between CIDR removal and TAI was higher in ECP group than in PGF-SC and PGF-IM groups (P < 0.001). Cows that displayed estrus has higher P/AI than cows that did not (P = 0.008). In conclusion, these results suggested that intramuscular or subcutaneous injection of PGF2α could be successfully used to induce ovulation in cattle undergoing TAI, with similar pregnancy rates when compared with ECP. The subcutaneous injection of PGF on the same day of CIDR removal could be an interesting alternative due it reduces cattle management to obtain similar results.

Antral follicular population (AFP) and anti-Müllerian hormone (AMH) concentration are strongly associated. Thus, analyzing AMH concentration is a reliable method to predict animals with high AFP, an important feature to select donor cows for embryos and oocytes. However, not much is known about this parameter in heifers, particularly in crossbred animals. The aim of this study was to assess AFP in heifers and cows and its relation to serum AMH levels. Bos taurus indicus cows (36-60-months-old; n = 30) and heifers (12-14-months-old; n = 17) of the same breed were evaluated. A single procedure of follicular counting was performed by ultrasound for each animal. Random sampling was performed to analyze serum AMH concentration. Cows showed higher AFP than heifers; nonetheless, plasma AMH concentration did not differ between the categories. A high correlation of AFP with plasma AMH concentration was observed in Girolando heifers. Thus, it is suggested that AMH concentration could be a reliable and less invasive method for selecting heifers with high AFP.

Dietary rumen-protected polyunsaturated fatty acids (PUFAs) rich in linoleic acid (LA) may affect embryo yield, and LA can modulate the molecular mechanisms of lipid uptake in bovine blastocysts produced in vitro. In embryos, membrane lipids, such as phosphatidylcholines (PCs) and sphingomyelins (SMs), affect cryopreservation success. The aim of the present study was to evaluate embryonic developmental rates after the IVF of oocytes retrieved from Nellore heifers fed for approximately 90 days with rumen-protected PUFAs rich in LA. In addition, we evaluated embryo cryotolerance and the membrane structure lipid composition using matrix-assisted laser desorption ionisation mass spectrometry of fresh and vitrified embryos. Embryo development to the blastocyst stage (mean 43.2%) and embryo survival after vitrification and warming (mean 79.3%) were unaffected by diet. The relative abundance of one lipid species (PC ether (PCe; 38:2, which means that this lipid has 38 carbon atoms and 2 double bonds in the fatty acyl residues) was increased after PUFAs supplementation. However, 10 ions were affected by cryopreservation; ions consistent with PC 32:0, PC 34:1, SM 24:1, PC 40:6 or PC 42:9, PC plasmalogen (PCp) 44:10 or PC 42:7, triacylglycerol (TAG) 54:9 and a not assigned ion (m/z 833.2) were lower in blastocysts that survived to the cryopreservation process compared with fresh blastocysts, whereas the abundance of the ions PC 36:3 or PC 34:0, PCe 38:2 or PC 36:6 and PC 36:5 or PCe 38:1 were increased after cryopreservation. Thus, the results demonstrate that the mass spectrometry profiles of PC, SM and TAG species differ significantly in bovine blastocysts upon cryopreservation. Because the lipid ion abundances of fresh and vitrified-warmed embryos were distinct, they can be used as potential markers of post-cryopreservation embryonic survival.

The aim was to compare the effects of two production systems on performance, carcass traits and physical-mechanical characteristics of leather from Beefalo-Nellore steers and heifers and to determine if the response to the production system was similar for both genders. A total of 40 Beefalo-Nellore cattle, 20 steers and 20 heifers, were evaluated. Animals were divided into two production systems: slaughtered at 15 (intensive system) or 26 (extensive system) months of age. In the intensive system, all animals received a ration containing 600 g/kg corn silage and 400 g/kg concentrate. In the extensive system, animals were kept on a pasture predominantly based on Brachiaria sp. and supplemented with 2 kg/day concentrate. In the intensive system, there was no difference in slaughter weight (470 kg body weight) between steers and heifers but steers in the extensive system had greater slaughter weight than heifers (463 and 428 kg body weight, respectively). Leather weight was higher for animals in the intensive than extensive system but there was no difference in leather weight once excess fat was removed. Leather quality from Beefalo-Nellore cattle slaughtered at 15 or 26 months of age is similar although carcass yield is higher for cattle slaughtered at a younger age.

This study evaluated the use of cooled semen in a fixed-time artificial insemination (FTAI) program compared with frozen-thawed semen to improve pregnancy rates in beef cattle. Ejaculates of three bulls were collected and divided into two treatments: (1) frozen-thawed semen and (2) cooled semen. Egg-yolk extender without glycerol was used for the cooled semen treatment. Straws (25×106 spermatozoa) were submitted to cooling for preservation at 5°C for 24h, after which FTAI was performed. Nelore cows (n=838) submitted to FTAI were randomly inseminated using frozen-thawed semen or cooled semen. There was a 20% increase in the pregnancy per AI (P AI-1) using cooled semen compared with frozen-thawed semen (59.9±4.7 vs 49.4±5.0%; P<0.005). There was no difference in P AI-1 among the bulls (P=0.40). The frozen-thawed semen had fewer functional spermatozoa than did the cooled semen when evaluated by sperm motility (61.7 vs 81.0%), slow thermoresistance test (41.7 vs 66.7%) and hypoosmotic swelling test (38.3 vs 53.7%; P<0.05). The percentage of sperm abnormalities did not differ between the freeze-thawing and cooling processes (18.6 vs 22.1%; P>0.05). Because there was less damage to spermatozoa and improvement in P AI-1, the use of cooled semen instead of frozen-thawed semen is an interesting approach to increase reproductive efficiency in cattle submitted to a FTAI protocol.

This study examined the effects of antioxidant supplementation and O2 tension on embryo development, cryotolerance and intracellular reactive oxygen species (ROS) levels. The antioxidant supplementation consisted of 0.6 mM cysteine (CYST); 0.6 mM cysteine + 100 µM cysteamine (C+C); 100 IU catalase (CAT) or 100 µM ß-mercaptoethanol (ß-ME) for 3 or 7 days of in vitro culture (IVC). Two O2 tensions (20% O2 [5% CO2 in air] or 7% O2, 5% CO2 and 88% N2 [gaseous mixture]) were examined. After 7 days of antioxidant supplementation, the blastocyst frequencies were adversely affected (P < 0.05) by CYST (11.2%) and C+C (1.44%), as well as by low O2 tension (17.2% and 11.11% for 20% and 7% O2, respectively) compared with the control (26.6%). The blastocyst re-expansion rates were not affected (P > 0.05) by the treatments (range, 66-100%). After 3 days of antioxidant supplementation, the blastocyst frequencies were not affected (P > 0.05) by any of the antioxidants (range, 43.6-48.5%), but they were reduced by low O2 tension (P < 0.05) (52.1% and 38.4% for 20% and 7% O2, respectively). The intracellular ROS levels, demonstrated as arbitrary fluorescence units, were not affected (P > 0.05) by antioxidant treatment (range, 0.78 to 0.95) or by O2 tension (0.86 and 0.88 for 20% and 7% O2, respectively). The re-expansion rates were not affected (P > 0.05) by any of the treatments (range, 63.6-93.3%). In conclusion, intracellular antioxidant supplementation and low O2 tension throughout the entire IVC period were deleterious to embryo development. However, antioxidant supplementation up to day 3 of IVC did not affect the blastocyst frequencies or intracellular ROS levels.

The effects of intracellular (cysteine and ß-mercaptoethanol) and extracellular (catalase) antioxidant supplementation at different times during in vitro production (IVM and/or in vitro culture (IVC)) on bovine embryo development, intracellular reactive oxygen species (ROS) levels, apoptosis and re-expansion rates after a vitrification-thawing process were examined. Blastocyst frequencies were not affected by either antioxidant supplementation (40.5%-56.4%) or the timing of supplementation (41.7%-55.4%) compared with control (48.7%; P>0.05). Similarly, antioxidants and the moment of supplementation did not affect (P>0.05) the total number of blastomeres (86.2-90.5 and 84.4-90.5, respectively) compared with control (85.7). However, the percentage of apoptotic cells was reduced (P<0.05) in groups supplemented during IVM (1.7%), IVC (2.0%) or both (1.8%) compared with control (4.3%). Intracellular ROS levels measured in Day 7 blastocysts were reduced (P<0.05) in all groups (0.60-0.78), with the exception of the group supplemented with ß-mercaptoethanol during IVC (0.88), which did not differ (P>0.05) from that in the control group (1.00). Re-expansion rates were not affected (P>0.05) by the treatments (50.0%-93.0%). In conclusion, antioxidant supplementation during IVM and/or IVC reduces intracellular ROS and the rate of apoptosis; however, supplementation does not increase embryonic development and survival after vitrification.
